Sample preparation


* The sample containing chromophores should be solubilized and as pure as possible.

* Ultracentrifugation of the sample prior to the measurements might be needed in order to remove aggregates and to avoid experimental artifacts.

* A minimum volume of 40 microL is needed for measurements (we have 4.10E-2, 0.4, 1.5 and 2 mL cuvettes available in the lab).

* Be aware that the system is very sensitive (it can work down to femtomolar). Working with too highly concentrated samples might lead to reabsorption processes and/or damaging of the PMT detector.
