* Export your data in 3 columns XY ASCII text format (excitation wavelength ; ellipticity signal ; PMT voltage).
Upper image : the 3 columns of an ascii file after exportation.
* If you use an Apple computer, this Applescript will automatically convert your ellipticity values into mean residue ellipticities. It assumes that Microsoft Excel and TextWrangler are installed on your system.
(Prior to the treatment, you can use this Applescript to automatically remove the extension of several ascii files)
* Mean residue ellipticities are calculated with the following formula : [Theta] = Theta / ( 10 x N x l x c ), where Theta is the observed ellipticity signal, N is the number of peptide bonds, l is the cuvette pathlength and c the concentration of the sample.
The script will also generate a second file (with a ".DIC" extension) that is compatible with Dicroprot for secondary structure analysis.
